Preliminary study on ISSR analysis and classification of wild Musa germplasm in Guangxi, China  [2011]

Inter simple sequence repeat (ISSR) molecular marker technology was used to assess the genetic diversity and classification of 27 wild banana accessions collected in Guangxi, China. ISSR analysis showed that out of 100 randomly chosen primers, 16 primers were polymorphic for the studied germplasm and generated 168 stabilised amplified bands of which 84.5% (142 bands) were polymorphic. Based on a data matrix and unweighted pair-group method of arithmetic average (UPGMA) clustering analysis, the classification results showed that, at a similarity level of 0.67, the studied accessions (27 accessions collected from Guangxi, two original wild species Musa acuminate and Musa balbisiana and four cultivated genotypes of the AAA, AA and ABB genome) could be classified into six groups, i.e. the Longzhou wild germplasm group, the Musa acuminate group, the Musa balbisiana group, the Guangxi wild germplasm group, the ‘Guangfen1’ (ABB) cultivar group and the ‘Dajiao’ (ABB) cultivar group. The wild germplasm collected in Guangxi showed a closer relationship to the ABB group ‘Dajiao’ and ‘Guangfen1’ than to the M. acuminate or M. balbisiana group. Obvious regional relationships among the collected germplasm suggested diverse origins and evolution paths of banana in Guangxi. However, the genotype of wild banana in Guangxi still needs to be further characterised by molecular and morphological tools.
