Hypoglycemic and repaired DNA damage effects of flavonoids of Ipomoea batatas leaves on type Ⅱ diabetes mellitus mice model [2012]

Hypoglycemic effect of flavonoids of Ipomoea batatas leaves (FIBL) and its effect on the repair of DNA damage of diabetes mellitus were studied. The high sugar and high fat diet and streptozotocin (STZ) induced typeⅡdiabetes mellitus (non-insulin dependent diabetes mellitus, NIDDM) mice model were used, and then divided into model control group, FIBL low-dose group, FIBL high-dose group and rosiglitazone group. Some normal mice were taken as normal control group. Mice of model control group and normal control group were daily fed 0.2 mL dH2O, respectively; mice of FIBL low-dose group and high-dose group were daily fed 75 and 150 mg/kg FIBL respectively and mice of RT were daily fed 3 mg/kg rosiglitazone, all for 28 d continuously. Observation of animals in general was carried out, and blood of tail vein was collected to measure glucose after 0, 7, 14, 21, 28 d. On 29 d, blood of heads was collected to measure lipids and lymphocytes were separated, and then single cell gel electrophoresis (SCGE) was used to test DNA damage. After the animal models had been successfully set up, NIDDM mice showed some symptoms, including polydipsia, polyuria, polyphagia and a certain degree of insulin resistance. Treatment results of FIBL showed that FIBL could control NIDDM mice from increasing body weight, decreasing blood glucose and blood lipid levels. FIBL could repair DNA damage of NIDDM mice.
