The present work was carried out to assess the antioxidant activity of virgin olive oil by the well known DPPH free radical scavenging method used for measuring the Antiradical Activity (A sub(AR)) and a recently developed Co(II)/EDTA-induced luminol chemiluminescence method used measuring the Hydroxyl Free Radical Scavenging Activity (SA sub(HFR)). The second method has been applied for the first time to measure the SA sub(HFR) of the methanolic fraction of virgin olive oil in the framework of this study. The antioxidant activities of 22 olive oils from two crop years, comprising seven different varieties and thirteen distinct geographic regions were studied: (13) extra virgin, (7) virgin, one lampante and one rancid olive oil. The induction of the samples were also measured by the Rancimat test to determine the resistance of olive oil samples to oxidation. An antiradical activity (A sub(AR)) assessment was made of both the olive oils and the methanolic extracts. The A sub(A) of the total fractions of olive oils varied from 55.12 to 278.75 mg caffeic acid equivalent (CAF)/kg of oil. The A sub(AR) values of the total fractions significantly correlated with both alpha-tocopherol (r = 0.8250) and total phenol (r = 0.7024) contents. It was proved that the antiradical activity of the total fraction of olive oil was mainly due to its alpha-tocopherol content. The A sub(AR) of the methanolic fractions of olive oils varied from 13.64 to 61.39 mg CAF/kg of oil. To
tal phenol contents were also significantly correlated (r = 0.7942) with the A sub(AR) of the methanolic fractions of olive oils. The SA sub(HFR) values of the methanolic extracts varied from 24.95 to 109.38 mg CAF/kg of oil. A strong correlation (r = 0.9178) was found between the SA sub(HFR) values of the methanolic extracts and the total phenol contents of the samples. The mean value of the results of SA sub(HFR) for the extra virgin olive oil (EVOO) and virgin olive oils (VOO) were 68.91 and 69.56 mg CAF/kg of oil, respectively. No significant differences were found between the mean values of the A sub(AR) for the EVOO and the VOO samples. In addition, the rancid oil showed considerable high antioxidant activity values according to the two antioxidant activity methods. In this study, Co(II)/EDTA-induced luminol chemiluminescence method was shown to be fast, reliable and useful method for assessing the antioxidant activity of the methanolic fraction of virgin olive oil.
