Objective of this research was to use the simplex method for obtaining the optimum condition of isoflavones extraction from defatted soybean with ethanol. The continuous double and single effect evaporators under vacuum pressure were used to remove ethanol from crude extract to produce soy molasses. The optimum condition of extraction was when using 1 kg of defatted soybean with 64 percent (v/v) ethanol of 151 at 63 deg C for 110 minutes per batch. The HPLC analysis of isoflavones in crude extract, soy molasses, and total amino in soy molasses showed 62.2340 mg/l, 219.3004 mg/l and 299.81 mg/dl respectively. From the study, the operating cost, excluded fixed cost, to produce isoflavones in soy molasses was 544.39 Bath/g. The analysis of chemical composition, heavy metal and aflatoxin B1 revealed the amount of 95.00 mg/dl of nitrogen, 593.75 mg/dl crude protein, 5.44 percent dry matter, 0.4230 ppm copper, 0.0005 ppm arsenic, which are still under toxicity limiting concentration, and no lead as well as no aflatoxin B1.
