Studies on the shoot-tip culture of apple, cv. "Fuji"

Tissue culture of 'Fuji' apple started from about 1.5 cm long shoot tips proliferated at a rate of 6.4 times in 4 weeks when grown on Murashige and Skoog (MS) medium with 3% sucrose, 0.1 mg/l gibberellic acid (GA3), 1.0 mg/l 6-benzylamino purine (BA) and indolebutyric acid (IBA). The best rooting was obtained on 1/4 MS medium supplemented with 3% sucrose and 1.0 mg/l IBA. Rooting percentage of shoot tips increased according to the increment of the concentration of sucrose added into 1/4 MS medium when cultured under light condition (16 hrs day-length for 6 weeks)
