Observation on somatic embryogenesis in the immature embryo culture of sunflower

1. The somatic embryogenesis was produced differently in the different cultivars of sunflower. 2. The higher concentration of sucrose made better somatic embryogenesis of immature embryos of sunflower. 3. Under the same condition, the immature embryos of 2mm in length had the highest percentage of somatic embryogenesis among the embryos with different lengths. 4. In the Nitsch mediums with 17.5% of sucrose and 0.5-10.0 ppm of zeatin, the percentage of somatic embryogenesis increased with the increase of zeatin supplemented in the immature embryo culture. 5. 2,4-D was able to make the embryos become the groups of embryonic cells, but was unable to make them differentiate. 6. Under microbcope, it could be observed that the immature embryos produced some embryonic groups of cells and some embryoids, many of which emerged from the cotyledon and hypocotyledonary axis, the cells surrounding the embryonic groups of cells degenerated, therefore there were some spaces between the embryonic groups of cells and the surrounding tissue.
