Growth responses of phalaenopsis "Toyland" seedlings affected by different light intensity and temperature

Phalaenopsis 'Toyland' seedlings propagated in vitro as new hybrid were grown in controlled growth chamber for 210 days in order to observe their responses to different temperatures and light intensities. The temperatures were maintained at 20/18, 25/23, and 31/29 deg C day/night, and the lights were 1.5, 7.0 and 22.0 Watt/m** (2), respectively. A combination of 25/23 deg C day/night temperature and 7.0 Watt/m** (2) light intensity resulted in maximum leaf growth of the orchid seedlings. Maximum fresh weight was observed at 25/23 deg C and light intensity of 7.0 Watt/m** (2), and the weight was decreased by increasing temperature. Sucrose content of the seedlings was accelerated by decreasing temperature and light intensity, and content of fructose and glucose was higher at 7.0 Watt/m** (2) at temperature of 20/18 deg C than 25/23 deg C or 31/29 deg C. Soluble fructose, glucose, and sucrose content were higher in roots than in leaves. the presence of purple spots in the leaf and purple coloration in the lower sides of the leaf was prominent when the seedlings were grown at low temperature by visual observation: however, no difference in flavonoid copigments composition in seedlings suggests that seedling population is very close to homozygous.
