Comparison of five different media for callus induction and plant regeneration of Indica X Japonica F2 plants

Four high callusing ability Japonica cultivars were crossed with six low callusing ability Indica cultivars and anthers from some of their F2 progenies grown during the Boro season were tested on five different media for callus induction and plant regeneration. The media included were modified potato medium-1(p1). Chaleff's R2, Gamborg's B5 N6 and H5. For callus induction both N6 and R2 performed better than the other three media. But plant regeneration of calli produced in P1 and R2 were higher. In both these media embryogenic callus production and direct regeneration have been observed. N6 is a good calli inducing medium especially for the japonicas but the regeneration ability of the calli from N6 is low. Further modification of the R2 and P1 media would be tried to obtain high callus induction and plant regeneration of, not only the Indica X Japonica types, but also of the pure Indicas.
