Identification of apple cultivars (Malus domestica Borkh.) by electrophoretic isozyme analysis

20 apple (Malus domestica Borkh.) cultivars were analysed by peroxidase (E.C.1.11.1.7), aspartate aminotransferase (E.C.2.6.1.1), 6 phospohogluconate dehydrogenase (E.C.1.1.1.44), malate dehydrogenase (E.C.1.1.1.37) and glucose phosphat isomerase (E.C.5.3.1.9) on starch gel. Existing electrophoretic methods were adapted to obtain satisfactory separation of enzymes. several protein extraction buffers and procedures were evaluated to optimize enzyme activity. Cultivars had different number and relative mobility of electrophoretic bands. The most useful isozyme system for distinguishing among the cultivars were peroxidase and 6-phosphogluconate dehydrogenase. 20 cultivars were classified into 17 peroxidase and 8 6-phosphogluconate dehydrogenase phenotypic groups. Stability of isozyme phenotypes was independent of sample timing, rootstock and location.
