Detection of potato leafroll virus key immunosorbent electron microscopy

Potato leafroll virus (PLRV) in infected potato stems and leaves, Physalis floridana and Datura tatula was detected by immunosorbent electron microscopy (ISEM). The number of PLRV particles per standard area (SA=1000 micron sq. m) in potato stems, leaves and sprouts and dormant tubers were 71364, 55909, 2273, and 909, respectively. In stems and leaves of P. floridana this was 2273 and 15455, respectively. The maximum dilution of potato stem and leaf extracts up to which PLRV could be detected was 1/32 and 1/8, respectively. Sensitivity of ISEM was four hundred times higher than that of the common method of electron microscopy; it was a rapid, sensitive, direct and reliable diagnostic method.
