Determination of selenium in horse blood samples by HGAAS(Hydride Generation Atomic Absorption Spectrometry) and EAAS (Electrothermal Atomic Absorption Spectometry)

Determination of selenium in horse whole blood by hydride generation and electrothermal atomic absorption spectrometry were described. A simple hydride generator was used. Some parameters affecting the measurement of selenium by both techniques were studied. Hydride generation followed by atomic absorption spectroscopy was a suitable method for routine analysis of selenium in 1.0-mL volume of horse whole blood. Blood samples were digested with a nitric-perchloric acid mixture, and the digested samples were analyzed for selenium by conversion into the hydrides by reaction with sodium borohydride in dilute hydrochloric acid, followed by measurement with atomic absorption spectrometry in an air-acetylene flame. The detection limit of the method was about 49 ng selenium per 1 ml. blood. Effects of the presence of some cations and anions on the determination of selenium with hydride generation technique were investigated. The use of standard addition method was recommended. The analytical results obtained for samples of NBS no.1566 oyster tissues and NBS no.1577 bovine liver agreed well with the certified values. Recoveries of selenium added to the digests of blood samples averaged 113 % with a relative standard deviation of 3.9 %.
