A research was carried out to study how the seed populations and their distribution over the soil profile change on a prairie subject to tillage at Santa Rosa experimental station, Valdivia (Chile). Evaluation of the seed distribution and movement was based on the number of seeds present at the different soil depths. Seed samplings were taken from five dephts in five dates in the tillering, identifying and accounting the species. A factorial variance analysis with a block random design, and diversity index and frequency was made. From this study it was concluded that the amount of seeds and species increases by passing the disc plow as a second running, but they decreased in depth. Significant interactions were observed in the deep treatments, which with an increment in number of soil tillage make more homogeneous the seed distribution on the soil profile. Species with higher contribution in the seed bank were: Leontodon nudicaulis, Plantago lanceolata and Trifolium repens. Most species of the seed bank showed a low presence percentage, indicating a high heterogeneity of the community
