Effects of fixation on immunohistochemical demonstration of Pasteurella haemolytica antigen

Pneumonic lesions induced by Pasteurella haemolytica, in 84 mice, were divided into 4 groups and fixed with 10% neutral buffered formalin, 10% formalin, Bouin's fluid and Carnoy's fluid, respectively. Immunoperoxidase technique was applied to detect the bacterial antigen in the lung tissue. The number of macrophages with bacterial antigen in the cytoplasm of a limited area was counted and compared with each fixative groups statistically. Bouin's fluid showed the best results to detect P. haemolytica antigen, while 10% neutral buffered formalin and 10% formalin showed the better results than Carnoy's fluid. In addition, tissue section was treated with proteinase. No efficiency for detection was observed with proteinase treatment after fixed with each fluid
