Biochemical studies on the nature of Orobanche germination stimulant(s) isolated from flax seed diffusates

A certain number of physiological chemical properties of natural non-host flax stimulant(s) was investigated in relation to faba bean host stimulant(s) plants. Three active bands in thin layer chromatography from original ether extracts of faba bean roots and flax seed diffusates, were varied in Rf values, under UV light as well as being varied in their stimulatory effects on a number of Orobanche spp. (Orobanche crenata, O. ramosa and O. aegyptiaca) in vitro. Flax stimulant(s) was soluble in highly organic solvents (petroleum ether, hexane, benzene, and ether), whereas, faba bean stimulant(s) was soluble in benzene and ether only. Natural Orobanche stimulant(s) produced by the host and non-host plants were stable in alkaline (NaH Co3 5 (%)) solutions, however, the alkaline media gave some decline in germination activity (19.6 (%)) of flax stimulant(s), coupled with moderate reduction in the germtube length of O. crenata seed in vitro. Stability at different temperatures (-20 up to 60 degree centigrades) showed no inhibitory effect on the biological activity of dry Orobanche germination stimulant(s) of both plants. Partial purification might indicate that there are, at least more than one natural Orobanche stimulant(s) which exists in faba bean roots and flax seed exudates, however, distinct differences in their biochemical properties might reflect unsimilarity in their chemical structures
