Application of tissue and cell culture techniques in developing productive triticale

The embryo rescue technique is an established method for production of primary triticale forms. Tissue and cell culture techniques can be applied for induction of somaclonal variation especially in terms of point mutations. Point mutations can be used in the preselection procedure of characters which are coded by single or only few genes. First of all, it is necessary to clarify the molecular biological mechanisms of somaclonal variation. In triticale, useful experiments of gene manipulation can only be induced if two conditions are fulfilled: The sufficient production of regenerated adult fertile plants from isolated cell material has to be ensured. Genes which are important in the field of plant breeding have to be localized and characterized by means of biochemical and molecular biological methods. In triticale the application of biotechnological methods like tissue or cell culture and gene manipulation is still at the initial stages. In future these modern techniques may complete the methodical instrumentation of plant breeding in a valuable manner, but will not replace the conventional methods of applied genetics
