A modified culture technique for graft formation in explanted internodes in vitro: its development and advantages [in United Kingdom]

In three leguminous species, Glycine max, Pisum sativum and Vigna sinensis, in vitro grafting succeeded effectively following the application of indole-3-acetic acid to the physiological apex of the vertically cultured, grafted internodes. Auxin resulted in vascular differentiation and formation of large number of xylem connections across the graft union. A basal application of the same in addition to the apical application, however, did not show any significant change in such graft development in these species. This led to the development of a modified and simplified culture technique for in vitro graft formation in explanted internodes. This technique has been found to be advantageous over the previous system.
