Serological diagnosis of ovine brucellosis: A comparison between serological findings and isolation of Brucella in sheep

Blood serum and selected lymph nodes of 506 ewes, 2 to 7 years old, were collected in slaughterhouses of Northern Greece, where 75% of the lambs are vaccinated with Rev. 1 each year. Of the 506 sera tested, positive reactions were obtained in 90(17.8%) by the complement fixation test (CFT), in 74(14.6%) by the Rose Bengal test (RBT) and in 33(6.5%) by the serum agglutination test (SAT). Of the 506 lymph node pools, Brucella melitensis was isolated in five (1%). The organism was isolated from (4.4%) of the CFT positive and (0.2 %) of the CFT negative animals, from (4.1 %) of the RBT positive and 0.5 of the RBT negative animals and none of the SAT positive and (1.1%) of the negative animals. Of the five culture positive animals, 4 were revealed as infected by CFT, 3 by RBT and none by SAT. Factors that may have contributed to this discrepancy between serological and bacteriological findings are discussed.
