Morphology and distribution of langerhans cell in the epidermis of cattle

In the histological sections stained with ZIO and AlPase methods the morphology and distribution of Langerhans cell (LC) in the epidermis of cattle were observed under light and electron microscopes. The results obtained were as follows. 1) langerhans cells (LCs) were generally localized at the low suprabasal layers of the epidermis with slight regional variations and were clearly distinguished from melanocytes by the AlPase-Dopa method. 2) LCs have numerous dendrites ranging from 1 to more than 15. The dendrites were always projected toward the stratum granulosum and some of them were penetrating into the stratum corneum especially at the hair follicular part of the epidermis. 3) LCs at the epidermis of the extremities tend to have smaller cell bodies but their dendrites are long. While, LCs at the neck and other areas with relatively thin epidermis have large cell bodies but dendrites are shorter than those of the extremities. The localization of the LC body and the thickness of the epidermis appeared to influence both the projection and form of LC dendrites. 4) The mean number of LCs per 1 mm2 of epidermis at 50 portions of the bovine skin was 182 ranging from 33 to 503 in one to three months old cattle and 649 ranging from 331 to 1394 in six months old cattle. The number of LCs increased with age. 5) by an electron microscope the characteristic ultrastructure of LC, such as numerous dendrites ramified, relatively clear cytoplasm, nucleus of irregular outline, Bribeck granules of tennis racket type and many cytoplasmic vacuoles was observed in the bovine epidermis. 6) The number of Birbeck granules within LC was 2.04+-1.32 in one to three months old cattle and 1.26+-0.43 in six months old cattle. 7) The strong activity of enzymes in the cell membrane and lysosome of LC was confirmed by electron microscopy
