Isolation of Brucella spp. from milk preserved of 15 deg C

Sixty milk samples preserved at -15 deg C for 20 months were tested. All samples were from seropositive cows and Brucella was isolated before freezing. After thawing the samples were cultured in the same selective medium and under the same conditions as initially. Brucella was isolated from 51 (85%) out of the 60 samples. These samples had formed more than ten colonies on initial inoculation. On the contrary, the nine negative samples had formed 3 to 5 colonies.
