Interspecific hybridization behaviour of selected isolates of Pleurotus species using conventional method

In monospore culture isolation of Pleurotus sp. Bhutan, P. florida and P. sajor-caju, direct spore suspension plating technique was employed. Hybridization was made between P. sp. Bhutan and P. florida; P. florida and P. sajor-caju and P. florida and P. florida. Visual evaluation based on mycelial growth appearance was made. It had been found out through this technique that the number of compatible strains exceeded the theoretical 25 percent ratio of positive reactions based on the tetrapolar heterothallic sexuality of certain Pleurotus species. This could be attributed to the lesser accuracy of visual evaluation. Microscope examination of some slides revealed formation of clamp connections which indicates hybridization between crosses. Hybridization could indicate prospects of strain improvement of Pleurotus species and consequently produce agronomically superior strains. The number of slides examined however, is not sufficient to support the number of compatible and incompatible strains discovered through visual evaluation. Nevertheless, the fact that hybridization occurred remains as shown by the presence of clamp connections
