A technique of enzyme-linked immunosorbent assay (ELISA) for detecting canine antibody against Entamoeba histolytica was established. The condition of ELISA with the coating antigen at a concentration of 5 mug/ml, test serum diluted at 1:100 and the conjugate at a concentration of 1:3,000 (1.3 mug/ml) was the optimum as determined by checkerboard titrations. Satisfactory specificity of E. histolytica antigen against the sera from dogs infected with various species of the protozoa and helminth was demonstrated. No cross-reactivity was found by the ELISA in anti-E. histolytica antibody from the immunized dog against various species of parasitic antigens
