Effects of sucrose concentration and medium pH on the formation of callus and shoot of in vitro culturing leaf explant in Africam violets [Saintpaulia ionantha]

The effects of the sucrose concentration and medium pH on callus formation, growth and shoot formation of leaf explants cultured in the African violet (Saintpaulia ionantha cv. Susie) were studied in vitro. The media consisted of modified Murashige and Skoog (MS) medium containing 0, 10, 30, 50 and 70 g/liters of sucrose in Expt.1 or with adjusted to 4.3, 4.8, 5.3, 6.3 and 6.8 medium pH value in Expt.2. The leaf tissue explants were cultured 6 weeks on an incubator maintained at 25 +- 2 degrees C under a cool-white fluorescent lamp (3.0 Klux) a 16-hr photoperiod. The number of days from leaf explants placed into solid medium to the beginning of callus formation and shoot formation was the shortest in culture containing 30 g/liter of sucrose and medium pH 5.8. The number of days from callus formation to shoot induction was shortened by higher concentration or more acidify. The formation percentage of callus and shoots, the shoots proliferated from the callus were maximized in cultures with containing 30 g/liter sucrose and medium pH 5.8. Total fresh weight of the formed callus and shoots regenerated from the callus were heaviest in cultures containing 30 g/liter sucrose and medium pH 4.8. The morphogenic by regenerated calli and shoots from employed explants in leaf tissue culture showed better results in leaf blade than those leaf petiole section
