Assessment of the response of Douglas fir embryocultures derived from various seed lots

Introduction of the Douglas fir Pseudotsuga menziesii (Mirb.) Franco to Slovak forests has a long tradition. On the other hand, there are several difficulties. One is that the quantity of Douglas fir seed from highly valuable domesticated stands is negligible. Priority has been given to the import of seed from unverified sources in the USA and Canada because of lower price as a criterion for purchasing seed lots. The aim of our work was to compare in vitro growth of embryos originating from Washington with seed from a "domesticated" subpopulation at Dobra Niva. Percentage of calogenesis, organogenesis and average number of adventitious buds in isolated embryos were observed and evaluated. Embryos were cultivated on MS medium supplemented with different concentrations of cytokinine 6-BAP. Transfer of explants to the fresh media was accomplished at regular intervals. No statistically significant differences between "domesticated" seed and the purchased seed lot from Washington was found. Calogenesis was on a similar level. The percentage of organogenesis was 28.96 % in the domestic provenance and 31.71 % in the seed from Washington. The average number of adventitious buds was 1.63 and 1.65, respectively. As to the micropropagation method, it is noted that induction of adventitious buds seems to be a suitable way for commercial propagation of valuable seed material originating from domestic sources
