Analysis of protein changes during the development of mung bean (Vigna radiata (L.) wilczek) seeds

Mung bean (Vigna radiata (L.) cv. Kampangsaen II) seed development was studied from 2 to 24 days after flowering (DAF). The seeds with seed wight of 0.870 gm reached the physiological maturity at the 16th DAF. Protein contents accumulated in cotyledons of mung bean embryos aged 2-24 DAF were determined. The total protein content changed slightly at beginning (2-8 DAF) and increased sharply from 8 to 16 DAF. The seed reached a maximal content of 152 mg/cotyledon pair at 16th DAF. Then the content declined slightly until to 24 DAF. The protein contents in epicotyl and hypocotyl of embryos developed from 10 to 20 DAF were also measured. The similar protein changes were observed in epicotyl and hypocotyl of embryos during this time. The developmental patterns of proteins expressed in cotyledon, epicotyl and hypocotyl of embryos were analyzed by SDS-gel electrophoresis. The expression of proteins in cotyledon started at 2th DAF and had a great changes during from 8 to 10 DAF, because there were 14 proteins synthesised during this time. The protein pattern remained unchanged from 16 to 24 DAF. The similar developmental patterns were observed in epicotyls and hypocotyls. The epicotyl and hypocotyl proteins expressed at the 10th DAF and had a little changes during 16 to 20 DAF. By 2 D-eletrophoresis, the cotyledon proteins have been identified into two groups: The first group consisted of proteins which were synthesised during the time course of development and then disappeared at later stages. The second ones were proteins which expressed and still remained through the time of physiological maturity. These were protein 21.7, 27, 48.7, 51, 54.6, 58 and 60.8 kD and would probably be storage proteins in mung bean seed cv. Kampangsean II.
