Initiation of root and callus cultures of Derris elliptica and D. philippinensis

Derris sp. roots, where rotenone is mostly concentrated, were induced from stem cuttings and propagated in vitro. Root explant was initially cultured in Murashige and Skoog (MS) medium supplemented with kinetin and 2,4 dichlorophenoxyacetic acid (2,4-D). Subsequently, a month-old root calli were subcultured onto MS medium containing kinetin and different levels of 2,4-D, and Woody Plant (WP) medium enriched with varying levels of 6-benzyladenine (BA) and 2,4-D. Callus formation was more rapid on WP medium with BA and 2,4-D than on MS medium with kinetin and 2,4-D. WP medium supplemented with the combination of 2 ppm BA and 10 ppm 2,4-D promoted highest callus growth rate. Callus multiplication was relatively faster when kept in total darkness. The growth of roots in a hydroponic system using modified Johnson's solution was also explored. Preliminary results indicate good root growth with lateral root formation
