Preliminary biochemical study of Phytophthora palmivora (bud rot) disease of coconut

Chemical analysis showed differences between healthy and bud rot infected samples. In general, infected samples had lower moisture content and free amino acids in aft, ash, crude fiber and nitrogen free extract (NFE) as well as in total phenols. The infected samples had a different color reaction (purple to dark blue vs. green) than the healthy samples. The infected samples consisting of the leaf base number 6 had more secondary metabolites such as coumarins and phenolic substances than the healthy samples. Few bands of terpenoid substances with little or no differences between healthy and infected samples were detected. Inoculation of seednuts with free culture of bud rot and nut rot isolates produced the following dissusates which hardened after 2 to 3 days brown patch around sites, browning inside husk, hydrolyzed endosperm, cracked shell causing leakage of water. The Malayan Red Dwarf (MRD) seednut was more sensitive than the green dwarf. Thin layer chromatography of ethanolic extracts of inoculated sites of the husk showed marked differences between control and culture filtrates unlike the results obtained in naturally infected bud
