Tissue cultures of silver fir and their defense reactions

The genus Abies is one of the largest groups within the conifers including about 50 species. Its sensitivity to drought and other environmental stresses make it one of the most damaged tree species in the recent forest decline. Following environmental stress, trees are very frequently damaged by pathogens. In attempting to develop a suitable regeneration system for the damaged species, we have concentrated on the development of suitable methods of Abies sp. micropropagation. In this study embryogenic and normal calli were induced from the same seed of Abies alba and their defence reactions were compared. Phaeolus schweinitzii (Fr.) Pat. wood-rotting fungus was used as a tester. In the presence of embryogenic callus, inhibition of mycelial growth was stronger than in the presence of a normal callus. The data indicate that the defence reaction of embryogenic callus is very intensive, probably reflecting the metabolic specificity of this tissue, which is auxin-independent
