Ultrastructural and autoradiographical analysis of nucleologenesis in the early goat embryos

Early pre-implantation goat embryos were investigated by fine-structure morphology and autoradiogradiographic detection of (5-sup(3)H)-uridine incorporation. Complex nuclear bodies were already seen in early pronuclei. These methodologies ofter clear-cut evidence of the already fully-developed nucleolar structure as well as of nucleoplasm labelling only in 16-cell embryos. Nucleoplasm labeling which is supposed to detect the onset of embryonal transcription was absent in the pronuclear and the 2-to 4-cell embryos. The first (5-sup(3)H)-uridine incorporation was detected in the 8-cell stage nuclei nucleoplasm followed by nucleolar labelling. Based on this evidence, we found that in the development of the early goat embryo there is no striking difference in the timing of the shift in control from the maternal to embryonic genome in comparison with other ruminants
