Inheritance of Embryoid Formation ability in soybean (Glycine max L. Merr.)

Study on the inheritance of embryoid formation ability was conducted in 3 Thai recommended soybean cultivars. The cultivars 'Chiang Mai 60'(CM 60) and 'Sukhothai 1'(SK 1) produce a number of embryoids while 'Nakhon Sawan 1'(NS 1) hardly produces any embryoids. Four crosses were made among them to obtain the F1 seeds of CM60 * NS1, NS1 * CM60, SK1 * NS1 and NS1 * SK1. Cotyledons were dissected from young seeds of F1 and F2 plants having the size of 3 to 5 mm (15 to 20 days after pollination) and cultured in an embryoid induction medium. The medium contains MS inorganic salts, vitamins from B5 medium, 7.5 mg/l NAA, 3 % sucrose, 0.65 % agar, and adjusted to the PH of 5.8. After culturing for 3 to 4 weeks the cotyledons from the F1 seed gave number of embryoids falling between the 2 parents in all 4 experiments. The cotyledons from individual F1 plants do not differ in embryoid formation. The cotyledons from F2 plants gave number of embryoids that were normally distributed, revealing that the embryoid formation ability in these soybeans was quantitatively inherited with the genes acting mainly in additive manner. Thus improvement of this trait may be achieved through routine selection methods employed in self-pollinated species.
