Cross protection between strains of peanut stripe virus

To further confirm that symptom variants of PStV are strains of RStV and not related potyviruses, a cross protection test was conducted using the Thai isolates. One hundred plants of Tainan 9 peanut at two weeks old were inoculated with the mild strain T1. Subsequently, they were challanged by blotch (T8), necrotic (T6), stripe (T2), chlorotic ring-CP-N (T3) and the mild strain (T1) at 3 weeks after the first inoculation. At least 15 infected plants were challanged by each strain and 5 healthy plants were similarly treated to serve as controls. Inoculated plants were examined periodically to detect any variation in symptoms after the challanged inoculation. At 2-3 weeks, most of the healthy plants that were inoculated showed symptoms typical of each strain, while the preimmuned plants showed not sign of new symptoms. Inocula prepared from these plants when back inoculated to Tainan 9 peanuts or cowpea KC84R (only those that had been challanged by T3) induced only symptoms of the mild strain. These results clearly indicated that the mild strain T1 could cross protect Tainan 9 from infection by other strains. It also proved that all the isolates used in this study are strains of PStV and not serologically related viruses.
