Methods are described for detecting beta-lactoglobulin C and alphas1-casein A variant genes. In the former case, a Pst1RFLP was used as an initial screen, but it was found advisable to confirm the analyses by sequencing putative C variant genes. In the latter case, the amplification created restriction site (ACRS) method was used to detect the specific alphas1-casein gene point deletion which gives rise to A variants in a New Zealand herd. We believe that ACRS is a particularly powerful method for detecting variant genes not amenable to the standard RFLP analyses.
