Genetic transformation of rice (Oryza sativa L.) using potato proteinase inhibitor 2 (PIN 2) gene insect resistance

Susceptibility of rice plants to a number of insect pests such as striped rice stemborer (Chilo suppressalis Walker) and the common cutworm (Spodoptera litura Fab.) causes severe reduction in yield. Insect resistance can be improved by incorporating potato proteinase inhibitor 2 (Pin 2) gene in the rice genome through genetic transformation. Pin 2 has adverse effects in the growth and development of susceptible insects and has insecticidal property if expressed in high quantity. Genetic transformation using this gene is presently being conducted in the following rice varieties: IR72, PSB Rc 14, LX (new plant type) and IR43. Plasmid construct pTWa containing the Pin 2 gene driven by its constitutive promoter and the bar gene driven by CaMV35S promoter was introduced to immature embryos and embryogenic calli induced from mature embryos via the particle inflow gun. Calli resistant to a selection medium containing 5.0 mg/L Basta were transferred into plant regeneration medium. Regenerants were obtained from resistant calli induced from mature seeds. Molecular analyses of the regenerated plants are presently being undertaken
