Aseptic culture techniques for the propagation of narra (Pterocarpus sp.)

This study aimed to assess the applicability of tissue culture techniques in the propagation of narra (Pterocarpus sp.) using different composition of basal medium. Specifically, this study was geared towards the screening of the nutrient media best suited for in-vitro callus induction of Pterocarpus indicus and P. vidalianus. Based on the results obtained, the total weight gained by the explant of the two Narra species (Factor A) was insignificant. The result indicated that neither of the two species used responded differently to in-vitro propagation. For the total number of callus developed in the two species of narra in 30 days of aseptic culture, both species did not vary in their relative response to the different media of aseptic propagation. The most appropriate culture media suitable for the induction of callus from young leaf and shoot of both narra species was the Murashige and Skoog's (MS) medium. This positive response of the explant was attributed to the more complete nutritional composition of the MS medium compared to the other media which lacked two or more nutritional factor needed for the in-vitro propagation. The MS medium provided the nitrogen nutrient from the two sources namely potassium nitrate (KNO3) and ammomium nitrate (NH4NO3). Tissue culture therefore is recommended as a tool for rapid mass propagation. The technique is advantageous over the traditional one because of its ability to produce more plantlets
