Development of selection techniques for blast resistance in aromatic rice variety KDML 105 by using culture filtrate and tissue culture

Investigation on pathogenecity of culture filtrate (CF) of Pyricularia oryzae Cav. revealed that CF of the fungus played as primary determinant of blast disease. CF obtained from isolate Khon Kaen was the most virulence. Lesion size and symptom severity were increased by the increasing of CF concentrations. The response of rice tissue to CF has been studied in vitro. The increasing of concentrations would reduce number of shoot and height of shoot, percent death of shoot was increased, size of callus was reduced, percent of callus death was increased, green spot formation was reduced and percent of regeneration of callus was not significant. The highest percent of regeneration, 12.5 percent, was obtained at concentration of 2:9. The variation on some agronomic characteristics of plant derived from culturing in media containing CF of P. oryzae was studied. The variation of leaf size, plant height and blast resistant of plants derived from tissue culture was higher than plants which were not derived from tissue culture. Rice varieties had the difference response to CF at concentration of 2:9. The purification of CF stated that the substance which produced by P. oryzae was Tenuazonic acid and it can produce the similar symptom as produced by the pathogen. The result suggested that Tenuazonic acid was the pathotoxin and it played an important role of pathogenecity of blast disease.
