Response of several red clover genotypes on callus formation and plant regeneration in several media systems

Six genotypes (7-1-5-36, 7-1-5-43, C34TC, 8-2-16-5, 8-2-16-39, FLMR6-23-3) of red clover that previously had been determined to be good regenerators on B5 protocol were tested for plant regeneration capacity using six media protocols (callus induction - embryo induction - embryo/plant development) were used throughout these studies. No significant differences in callus fresh weight was observed among media, however genotype by media interaction was significant. Highly significant differences in plant regeneration were observed for genotypes, media, and for the interaction between genotype by media. In second experiment, ten genotypes of red clover (NEWRC1, NEWRC4, NEWRC30, NEWRC64, NEWRC74, FLMR6-23-3, C34TC, C54TC, 7-1-5-36) which have a range in levels of plant regeneration capacity on B5 protocol were used to compare three media systems chosen for their callus formation and plant regeneration capacity. No significant differences among media or genotypes for callus fresh weight gain at 4-wk, were observed. Varying levels of regeneration of the genotypes in different media indicates that there is a significant effect of plant genotype x media interactions

Enam genotip red clover (7-1-5-36, 7-1-5-43, C34TC, 8-2-16-5, 8-2-16-39, FLMR6-23-2) yang merupakan regenerator yang baik pada protokol media B5 diuji kemampuan beregenerasinya pada 6 sistem media. Protokol media tiga tahap (induksi kalus-induksi embrio-perkembangan embrio/plantlet) dilakukan pada seluruh penelitian ini. Tidak ada perbedaan yang nyata diantara media, walaupun demikian interaksi antara media dan genotip nyata. Perbedaan yang sangat nyata dalam regenerasi tanaman terdapat diantara genotip, media dan interaksinya. Pada penelitian yang kedua, sepuluh genotip red clover (NEWRC1, NEWRC4, NEWRC30, NEWRC64, NEWRC74, FLMR6-23-3, C34TC, C54TC, 7-1-5-36) yang mempunyai kisaran yang luas dalam tingkat kemampuan beregenerasi tanaman dalam protokol B5 digunakan untuk membandingkan media terpilih untuk pembentukan kalus dan kemampuannya beregenerasi. Tidak ada perbedaan yang nyata diantara media maupun diantara genotip dalam berat basah kalus setelah 4 minggu. Tingkat regenerasi tanaman yang berbeda pada genotip menunjukkan adanya pengaruh yang nyata pada interaksi antara media dan genotip
