Improved micropropagation of yam, Dioscorea rotundata in liquid cultures

Experiments were conducted toward developing an improved method for micropropagation of yam. The phytohormone regime and tissue support mechanism were varied in cultures on liquid media. Addition of 1 mg/l kinetin (kin) significantly increased the number of shoots and nodes produced and root length but caused a decrease in number of roots. Neither leaf size nor percent of leaves in the biomass were affected. Internodes were notably shorter in cultures grown in media containing kin. Flurprimidol (fp) in bud multiplication and rooting media for yam appears to have enhanced each of the processes which produce compact plants by limiting leaf expansion and petiole elongation. Maximum shoot multiplication was observed in media supplemented with 3-6 mg/l fp. Shoot multiplication and root formation were better on stationary membrane rafts than in shaken flasks
