Effect of NaCl on callogenesis of Triticum aestivum L.

Seeds of three cultivars of Triticum aestivum (evs. Lyallpur 73, Pak-81 and Hyderabad-88) were studied for callus induction on Murashige and Skoog medium containing 3 mg/I 2,4-dichlorophenoxy acetic acid (2,4-D) with and without varying concentrations of NaCI. Callus induction frequency increased with the addition of 50mM NaCI to the medium and then decreased with the increase of stress in the medium. Calli were well proliferated at 5OmM NaCI, while at higher salt concentrations their water content was lost and they represented degenerated tissue.
