Aujeszky's disease virus (SHV-1) challenge dose titration study

Study aims to determine the optimal intranasal (in. virulent virus (RICE strain) challenge dose of fattening pigs and 7 days old piglets for the immunogenicity study required in the European Pharmacopoeia (EP). The infection of fattening pigs (80-100 kg) by 10*[4), 10*[5), and 10*[6) infective dosis resulted Aujeszky's disease similar to observed in field outbreaks. On the basis of the values of the body weight changes all the three infected groups met the challenge of values required by the EP. In the immunization experiment the given test conditions decide, which challenge dose (10*[4), 10*[5), 10*[6) PFU/ml/nostril) will be used. The in. infection of 7 days old piglets by 10*[1), 10*[2) and 100[3) infective doses of virulent virus resulted Aujeszky's disease characterised by the CNS signs similar to observed in field outbreaks. On the basis of 100 percent mortality rate all the three infected groups met the challenge values required by the EP. Considering the unsatisfying effect of the infection with 10*[1) dose, in the immunization experiments the 10*[2) and 10*[3) minimal challenge dose of virulent SHV-1 can be suggested for in. infection of 7 days old piglet. The study was conducted according to GCP (good clinical practice). The experiment was approved by the decision of the committee for the Ethical Control of Animal Experiment.
