Use of HP-size exclusion chromatography to study the degree of polymerization of silk (Bombyx mori) fibroin fibres

The present study deals with the chromatographic determination of molecular weight and degree of polymerization of Bombyx mori silk fibroin by HP-Size Exclusion Chromatography (HP-SEC). The column was calibrated with three series of water-soluble molecular weight standards: proteins, pullulans, and polyethylene oxides. The effect of a denaturing mobile phase, containing 5M urea, on the elution behaviour of the standard polymers was investigated. The calibration curve of proteins was significantly affected by the presence of the chaotrope, while those of the two other polymers remained unchanged. Silk fibroin samples with different degree of polymerization were dissolved with saturated aqueous LiSCN and analysed by HP-SEC after dialysis against the mobile phase. The chromatographic profiles allowed the following parameters to be calculated,for each sample: Mw, (weight-average molecular weight), Mn (number-average molecular weight), Mw/Mn (polydispersity index), MWD (molecular weight distribution). The intrinsic viscosity [eta] of the samples was measured according to the SNV 195 595 standard method and a relation between the values of [eta] and Mw was established. The advantages of using the HP-SEC method for the determination of the degree of polymerization of silk fibroin fibres are discussed
