Strain characterisation of plum pox virus isolates by the use of monoclonal antibodies

A largen scale survey was carried out,on the basis of symptoms incidence and the results from serological testing for the elucidation of PPV sanitary status in relation with its strain characterization.DAS-ELISA with polyclonal antibodies and DASI-ELISA with two strain specific (M and D) monoclonal antibodies were used.Different types of symptoms expressed as rings,spots,mottling,stripes along the nerves, vein clearing and chlorosis were observed. ELISA results showed that PPV was common in the tested plum (P.domestica)cultivars, with 77.2% rate of infection.Unlike the plum cultivars,all tested P.cerasifera and P.intersp. hybrids cultivars and both cultivars from P.salicina were PPV free.PPV strain characterization and differentiation showed the presence and distribution of only PPV-M strain (100%).
