Development of a fermentation medium and detection bioassay for the local production of the anthelmintic antibiotic avermectins

Streptomyces avermitilis, the producer of the anthelminthic compound for animals, avermectin, was found to grow best and produce the highest yield on Dutton's seed and production media. Cheaper and locally available substrates were screened as substitutes for the carbon and nitrogen sources of the medium. Among cassava starch, all-purpose flour and cane sugar, cassava starch was found to be the best substitute for soluble starch. Baker's yeast was established to be a better substitute for cottonseed flour than soymean meal. Restricted random balance design experiments were performed on the modified production medium 8 (MPM8) to come up with an optimized formulation of MPM8 consisting of cassava starch, baker's yeast, potassium phosphate and zinc sulfate. To assay the production of avermectin, the brine shrimp Artemia franciscana was used. The Artemia cysts were hatched in saline solution incubated for 24 hours with continuous aeration and illumination. The larvae were incubated in 30-ml vials containing the avermectin solutions for two hours. The number of dead brine shrimps was counted and the % mortality was computed. Avermectin activity was estimated based on a standard curve established indicating the effect of known concentration of avermectin on the mortality of the brine shrimps
