Embryo culture, callogenesis and regeneration techniques in Pisum sativum L. in vitro

Studies on embryo culture, callogenesis and regeneration techniques in Pisum sativum L., in vitro were performed in Biotechnology Laboratory, Department of Plant Breeding and Genetics, Sindh Agriculture University, Tandojam, during 1995. Two exotic and a local variety of Pisum sativum L. namely Australian (V1), Swiss (V2) and local (V3) were used for embryo culture on MS. medium. The shoot tip of the plants obtained from embryo culture were again cultured on MS. medium in order to get regenerated plants. Better results were achieved in case of embryo culture as well as shoot tip culture in Australian variety (V1) as compared to Swiss (V2) and local (V3) varieties. Pieces of stem, embryos and leaves of all the three varieties were used as explant materials to establish callus on MS. medium with different compositions of 2, 4-D like 2, 3 and 4mg/l in addition to growth regulators like, Kinetin, IAA and IBA. It was observed that stem as explant material used from Australian variety (V1) established progressive callus when cultured on MS medium containing 3mg/l of 2, 4-D over rest of varieties. The success rate of regeneration was generally low, however, Australian variety (V1) proved better as compared to Swiss (V2) and local (V3).
