Tagging Lr19 with AFLP markers

Despite reports of virulence in leaf rust (Puccinia recondita Rob. ex Desm. f. sp. tritici) to wheats carrying Lr19, this gene, which originates from Thinopyrum sp., remains an excellent source of resistance against all predominant South African pathotypes. A physical deletion map of the Lr 19 translocated chromosome facilitated the selection and characterisation of a white endosperm recombinant (LrI9-149). To achieve the aim of further shortening the Thinopyrum segment present in LrI9-149, and for subsequent selection and tagging of novel recombinants, genetic markers are required which lie closer to Lr 19 than any of the presently mapped marker loci. The AFLP technique has been employed to identify DNA sequences mapping in close proximity to Lr19, and these sequences can be used to design PCR based assays. Such assays are well suited to marker-assisted selection strategies.
