Identification of the sugarcane grassy shoot causal agents by using double-stranded RNA method

Grassy shoot disease is found in the central plain area which no evidence for the causal agent has been reported. However, some papers reported the posssibility of the causal agent is the same as white leaf disease. Infact, symtoms of grassy shoot are more different and varier than white leaf disease such as, the excessive tillering the softening of green leaf texture and some mature cane may produce white or chlorosis leaf. The objective of this study is to determine whether the grassy shoot is infected with virus or virus-like agents by using double-stranded RNA method. Double-stranded RNA (dsRNA) extraction from the healthy and grassy shoot cane were analysed by polyacrylamide gel electrophoresis. The result showed that there was no dsRNA from the healthy but there were two district bands of 5.3x10*[6) and 1.6x10*[6) from grassy shoot sample when the white leaf had only one band of 1.6x10*[6). To prove that those bands of nucleic acid were dsRNAs, the extractions were treated with DNase and then the band of 1.6x10*[6) was removed from both samples. Finally, dsRNA of 5.3x10*[6) can only be detected from grassy shoot disease. It was remarkable that the extra DNA band was not normally found in dsRNA extraction. So it is possible to be the DNA from the causal agent rather than DNA from host and it shold be dsDNA or secondary structure.
