Tissue culture of Gloriosa superba L.

Gloriosa superba L. contain major alkaloid colchicine which is used in the treatment of gout. Presently, Gloriosa superba L. is an increase in the demand of raw materials for colchicine in industrial by government and private company. Technique of plant tissue and cell culture was applied for Gloriosa superba L. MS media contained BA 2 ppm and NAA 1 ppm has been used for culture of root culture. After 4 weeks, root culture was dried and extracted with ethanol 95 percent. Extraction was studied qualitative analysis by TLC and quantitative analysis by HPLC. Root culture was found colchicine and 3-DMC which its derivative. Colchicine concentration in root culture and tuber of Gloriosa superba L. were determined by HPLC showed 0.0419 percent and 0.1402 percent on the dry weight basis, respectively.
