Detection of Glu D1 5+10 locus in gene resources of wheat (Triticum aestivum) with non-standard spike morphotype using DNA markers

In selected gene resources of Triticum aestivum with non-standard spike morphotype MRS (multirow spike) and LG (long glumes) presence of Glu D1 5+10 glutenin subunits were tested using DNA markers. The presence of this locus in a genetic marker of high breadmaking quality. The DNA for analysis was isolated from young wheat leaves using DNAsy Plant Mini Kit. Specific sequences of nucleotides in amplified segments, which were determined by specific polymorphic amplification test - SPLAT, were used of primers JEDL11 and JEDL12. The allele Glu D1 5+10 verification in MRS lines is specified. A relatively lower grain weight in MRS lines seems to be accompanied by higher protein content in grain due to an increased proportion of aleurone layers in grain. The genotypes with very high protein content ar used for further crosses and breeding.
