Mapping and marker-assisted selection in Maize and Wheat at CIMMYT

Scientists in the Applied biotechnology Center at the International Maize and Wheat Improvement Center (CIMMYT) have been addressing the challenge of incorporating biotechnology tools for the improvement of two cereal crops, maize and wheat. Two areas of research, molecular genetics and genetic engineering, have been employed. In molecular genetics, we have used various molecular markers to study in genetic diversity of maize and wheat germplasm, and to map a variety of traits in both species. Marker assisted selection (MAS) for three distinct quantitative traits has been carried out in maize with varying levels of success. We are now routinely using markers to select for a modest number of qualitative traits, including the opaque2 allele in maize. In wheat, selection is routine for the Phlb deletation, a thinopyrum intermedium translocation conferring resistance to BYDV, and the Cre3 gene for resistance to the cereal cyst nematode. This paper will however focus on an example of mapping a complex trait in each crop and the consequence for MAS.
