Genetic analysis of low tiller in japonica type rice cultivar Aikawa 1

The genetic mechanism of low tiller number in Japonica type rice Aikawa 1 was clarified using molecular markers. The single dominate gene control was confirmed from the two kinds of segregation analyzed of backcrossed population BC1F1 (n=33 plants, normal: low tiller=17:16, square 2 = 0.03, P=0.86) and its progenies, BC1F2 (n=33 lines, normal homozygote: heterozygote=16:17, square 2 =0.03, P=0.86) derived from crosses between Aikawa 1 and IR64 as a recurrent parent. Whole genome chromosome survey were carried out fine the linked DNA markers with the low tiller number gene using 64 SSR markers and ten BC1F2 lines that segregated low tiller plants. A total of eight markers on chromosomes 4, 8, and 10 showed the skew segregation that almost had the heterozygote in each marker. Using these markers and additional RFLP markers, the genotypes of whole lines of 33 BC1F2 were investigated. The linkage among low tiller number gene and DNA markers were analyzed by computer software, Map-maker/EXP ver.3.0. And then several DNA markers on long arm of chromosome 8 were found to link with the gene. Particularly, a SSR marker, RM 264 linked most closely to the gene with the distance of 6.1 cM. Because no gene related with low tiller number was reported in this chromosome region, the gene was designated as Ltn (t), tentatively. Using the genetic information, the linked DNA markers and Ltn (t), the authors are developing some low tiller rice with the IRRI leading varieties, IR64 and IR72 genetic backgrounds to clarify the yield ability of low tiller plant type and provide the breeding material for Indica type rice
