How the storage of plant roots can influence the isolation of fluorescent Pseudomonads from the rhizosphere

The roots of onion, radish, cucumber and lettuce were incubated at room temperature and at 4-6 deg C for 24 hours, and then fluorescent pseudomonads were enumerated on S1 selective medium. The number of pseudomonads obtained from stored roots, were compared to the number of these bacteria isolated immediately after roots were removed from the soil. The incubation of onion and radish roots at room temperature for 24 h significantly increased the number of isolated colonies of the pseudomonads in comparison to immediate isolation from fresh roots. The storage of these roots in the fridge generally decreased the number of the bacteria or had no effect at all. The storing of cucumber and lettuce roots at room temperature as well as at 4-6 deg C, significantly decreased number of isolated pseudomonads
