Identification and determination of extracellular invertase in suspension cultures of carrot

A simple and rapid method of the determination of glucose was used for the identification of invertase (beta-D-fructofuranosidase, EC 3.2.1.26) as well as for the determination of its enzymatic activity. Culture medium, in which cell suspension cultures of carrot (Daucus carota L. cv. Kalisto) were grown, were used as a source of the extracellular fraction of the enzyme. Homogenized cells of the suspension culture were used as a source of the intracellular fraction. The amount of glucose released from the sucrose subtrate was determined. Distribution of the invertase activity was: 421 nkat.g-1 dry matter (88.6%) in the intracellular fraction and 52 nkat.g-1 dry matter (11.4%) in the extracellular fraction. The specific activity of the intracellular enzyme was 5.9-times higher than that of the extracellular one
